A study on rapid acid chrome black (MB 7) spectrophotometric determination of ClO

摘要: Experiments were conducted to investigate the degradation of 2,6-dinitro- -cresol (DNPC) in the chlorine dioxide (ClO ) catalytic oxidation process. Pure aluminum oxide was used as the catalyst in this process. The degradation of DNPC by ClO using aluminum oxide as catalyst was systematically studied by varying the experimental parameters, such as pH values, catalyst dosage, the initial concentration of DNPC and ClO , reaction time, etc. Under optimal condition (DNPC concentration 39 mg·L , ClO concentration 0.234 g·L , reaction time 15 min, catalyst dosage 4.7 g·L and pH 4.32), almost complete degradation of DNPC can be achieved. The kinetic studies revealed that the ClO catalytic oxidation degradation of DNPC followed pseudo-first-order kinetics with respect to both ClO and DNPC concentration. The repetitive use of the catalyst was investigated along sequential feed-batch trials. The catalyst performed efficiently after five runs. In addition, a simple and convenient method for the determination of ClO in water was developed by using acid chrome black 7 (MB 7) spectrophotometry in this paper.

Expression of STAT6 and NF-κB p65 in the colon mucosa of patients with ulcerative colitis

摘要: The expression of signal transducer and activator of transcription 6 (STAT6) and nuclear factor-κB (NF-κB) in the colonic mucosa of patients with ulcerative colitis (UC) was examined. Real-time polymerase chain reaction and immunohistochemistry were used to detect the expression of STAT6 and NF-κB p65 at both mRNA and protein levels in the colonic mucosa of patients with UC and healthy volunteers. The results showed that the expression levels of STAT6 and NFκB p65 in the colonic mucosa of patients with UC were significantly higher than in normal controls at both mRNA and protein levels. These data suggest that STAT6 and NFκB p65 perhaps play an important role in the pathogenesis of UC and underscore the potential value of anti-UC strategies in the clinical management of this disease.

Effect of compound Sophorae Flavescentis Jiechangrong capsule on expression of NF-κB p65 and STAT6

摘要: The effects of compound Sophorae Flavescentis Jiechangrong capsule (CSFJC) on the expression of nuclear factor-κB p65 (NF-κB p65) and signal transducer and activator of transcription 6 (STAT6) in the intestinal mucosa of patients with ulcerative colitis and the possible mechanism were investigated. Eighteen patients with ulcerative colitis were randomly divided into a traditional Chinese medicine (TCM) group ( = 11) treated by CSFJC and a western medicine (WM) group ( = 7) treated by Sulfasalazine tablets. The treatment duration lasted eight weeks. Before and after the treatment, the symptoms and the physical signs were observed, and the routine stool test, the colonoscopy, and pathological examination were performed in the two groups. The expression levels of NF-κBp65 and STAT6 were detected by using immunohistochemistry. The results showed that the total effective rate of the curative effectiveness in TCM and WM groups was 100% and 71.4%, respectively, and the total effective rate of colonic mucosa lesion in TCM and WM groups was 90.9% and 71.4%, respectively, with the differences being significant (all < 0.05). The total effective rate of syndromes of damp-heat blocking according to the TCM in TCM and WM groups was 90.9% and 71.4%, respectively. After the treatment, the expression of NF-κB p65 and STAT6 in the two groups was decreased, and the decrease of NF-κB p65 and STAT6 expression in TCM group was more significant than in WM group ( < 0.05). It was concluded that CSFJC can inhibit the activation and expression of NF-κB p65 and STAT6 in the intestinal mucosa of patients with ulcerative colitis, which is a possible mechanism for CSFJC treating ulcerative colitis.

Ascalable and efficient IPv4 address sharing approach in IPv6 transition scenarios

摘要: IPv6 has been an inevitable trend with the depletion of the global IPv4 address space. However, new IPv6 users still need public IPv4 addresses to access global IPv4 users/resources, making it important for providers to share scarce global IPv4 addresses effectively. There are two categories of solutions to the problem, carrier-grade NAT (CGN) and ‘A+P’ (each customer sharing the same IPv4 address is assigned an excluded port range). However, both of them have limitations. Specifically, CGN solutions are not scalable and can bring much complexity in managing customers in large-scale deployments, while A+P solutions are not flexible enough to meet dynamic port requirements. In this paper, we propose a hybrid mechanism to improve current solutions and have deployed it in the Tsinghua University Campus Network. The real traffic data shows that our mechanism can utilize limited IPv4 addresses efficiently without degrading the performance of applications on end hosts. Based on the enhanced mechanism, we propose a method to help service providers make address plans based on their own traffic patterns and actual requirements.

Predictive models for machining operations have been significantly improved through numerous methods in recent decades. This study proposed a 3D finite element modeling (3D FEM) approach for the micro end-milling of Al6061-T6. Finite element (FE) simulations were performed under different cutting conditions to obtain realistic numerical predictions of chip flow, burr formation, and cutting forces. FE modeling displayed notable advantages, such as capability to easily handle any type of tool geometry and any side effect on chip formation, including thermal aspect and material property changes. The proposed 3D FE model considers the effects of mill helix angle and cutting edge radius on the chip. The prediction capability of the FE model was validated by comparing numerical model and experimental test results. Burr dimension trends were correlated with force profile shapes. However, the FE predictions overestimated the real force magnitude. This overestimation indicates that the model requires further development.

Cytokine storm and translating IL-6 biology into effective treatments for COVID-19

摘要: As of May 3, 2023, the coronavirus disease 2019 (COVID-19) pandemic has resulted in more than 760 million confirmed cases and over 6.9 million deaths. Several patients have developed pneumonia, which can deteriorate into acute respiratory distress syndrome. The primary etiology may be attributed to cytokine storm, which is triggered by the excessive release of proinflammatory cytokines and subsequently leads to immune dysregulation. Considering that high levels of interleukin-6 (IL-6) have been detected in several highly pathogenic coronavirus-infected diseases, such as severe acute respiratory syndrome in 2002, the Middle East respiratory syndrome in 2012, and COVID-19, the IL-6 pathway has emerged as a key in the pathogenesis of this hyperinflammatory state. Thus, we review the history of cytokine storm and the process of targeting IL-6 signaling to elucidate the pivotal role played by tocilizumab in combating COVID-19.

FERM domain-containing protein FRMD6 activates the mTOR signaling pathway and promotes lung cancer progression

摘要: FRMD6, a member of the 4.1 ezrin–radixin–moesin domain-containing protein family, has been reported to inhibit tumor progression in multiple cancers. Here, we demonstrate the involvement of FRMD6 in lung cancer progression. We find that FRMD6 is overexpressed in lung cancer tissues relative to in normal lung tissues. In addition, the enhanced expression of FRMD6 is associated with poor outcomes in patients with lung squamous cell carcinoma (n = 75, P = 0.0054) and lung adenocarcinoma (n = 94, P = 0.0330). Cell migration and proliferation in vitro and tumor formation in vivo are promoted by FRMD6 but are suppressed by the depletion of FRMD6. Mechanistically, FRMD6 interacts and colocalizes with mTOR and S6K, which are the key molecules of the mTOR signaling pathway. FRMD6 markedly enhances the interaction between mTOR and S6K, subsequently increasing the levels of endogenous pS6K and downstream pS6 in lung cancer cells. Furthermore, knocking out FRMD6 inhibits the activation of the mTOR signaling pathway in Frmd6−/− gene KO MEFs and mice. Altogether, our results show that FRMD6 contributes to lung cancer progression by activating the mTOR signaling pathway.

N6-methyladenosine (m6A) is the most common post-transcriptional RNA modification throughout the transcriptome, affecting fundamental aspects of RNA metabolism. m6A modification could be installed by m6A “writers” composed of core catalytic components (METTL3/METTL14/WTAP) and newly defined regulators and removed by m6A “erasers” (FTO and ALKBH5). The function of m6A is executed by m6A “readers” that bind to m6A directly (YTH domain-containing proteins, eIF3 and IGF2BPs) or indirectly (HNRNPA2B1). In the past few years, advances in m6A modulators (“writers,” “erasers,” and “readers”) have remarkably renewed our understanding of the function and regulation of m6A in different cells under normal or disease conditions. However, the mechanism and the regulatory network of m6A are still largely unknown. Moreover, investigations of the m6A physiological roles in human diseases are limited. In this review, we summarize the recent advances in m6A research and highlight the functional relevance and importance of m6A modification in in vitro cell lines, in physiological contexts, and in cancers.

Antibacterial and anti-flaming PA6 composite with metathetically prepared nano AgCl@BaSO

摘要: In this study, a facile and environmentally friendly method with low energy consumption for preparing nanoscale AgCl and BaSO co-precipitates (AgCl@BaSO co-precipitates) was developed based on the metathetical reaction. Then, the dried co-precipitates were melt-compounded with polyamide 6 (PA6) resins at a specified mass ratio in a twin-screw extruder. The results demonstrated that in the absence of any coating agent or carrier, the nanoparticles of AgCl@BaSO co-precipitates were homogeneously dispersed in the PA6 matrix. Further analysis showed that after the addition of AgCl@BaSO co-precipitates, the antibacterial performance, along with the flame-retardance and anti-dripping characteristics of PA6, was enhanced significantly. In addition, the PA6 composites possessed high spinnability in producing pre-oriented yarn.

P2P energy trading via public power networks: Practical challenges, emerging solutions, and the way forward

摘要: Peer-to-peer (P2P) energy trading is an emerging energy supply paradigm where customers with distributed energy resources (DERs) are allowed to directly trade and share electricity with each other. P2P energy trading can facilitate local power and energy balance, thus being a potential way to manage the rapidly increasing number of DERs in net zero transition. It is of great importance to explore P2P energy trading via public power networks, to which most DERs are connected. Despite the extensive research on P2P energy trading, there has been little large-scale commercial deployment in practice across the world. In this paper, the practical challenges of conducting P2P energy trading via public power networks are identified and presented, based on the analysis of a practical Local Virtual Private Networks (LVPNs) case in North Wales, UK. The ongoing efforts and emerging solutions to tackling the challenges are then summarized and critically reviewed. Finally, the way forward for facilitating P2P energy trading via public power networks is proposed.

A systematic survey of LU domain-containing proteins reveals a novel human gene, LY6A, which encodesthe candidate ortholog of mouse Ly-6A/Sca-1 and is aberrantly expressed in pituitary tumors

摘要: The Ly-6 and uPAR (LU) domain-containing proteins represent a large family of cell-surface markers. In particular, mouse Ly-6A/Sca-1 is a widely used marker for various stem cells; however, its human ortholog is missing. In this study, based on a systematic survey and comparative genomic study of mouse and human LU domain-containing proteins, we identified a previously unannotated human gene encoding the candidate ortholog of mouse Ly-6A/Sca-1. This gene, hereby named LY6A, reversely overlaps with a lncRNA gene in the majority of exonic sequences. We found that LY6A is aberrantly expressed in pituitary tumors, but not in normal pituitary tissues, and may contribute to tumorigenesis. Similar to mouse Ly-6A/Sca-1, human LY6A is also upregulated by interferon, suggesting a conserved transcriptional regulatory mechanism between humans and mice. We cloned the full-length LY6A cDNA, whose encoded protein sequence, domain architecture, and exon‒intron structures are all well conserved with mouse Ly-6A/Sca-1. Ectopic expression of the LY6A protein in cells demonstrates that it acts the same as mouse Ly-6A/Sca-1 in their processing and glycosylphosphatidylinositol anchoring to the cell membrane. Collectively, these studies unveil a novel human gene encoding a candidate biomarker and provide an interesting model gene for studying gene regulatory and evolutionary mechanisms.

G protein-coupled receptor LGR6 is an independent risk factor for colon adenocarcinoma

摘要: LGR6 is a member of the G protein-coupled receptor family that plays a tumor-suppressive role in colon cancer. However, the relationship between LGR6 expression in patients and clinicopathological factors remains unclear. This study aimed to clarify whether the expression level of LGR6 is correlated with colon adenocarcinoma progression. Immunohistochemistry was used to detect LGR6 expression in colon adenoma tissues ( = 21), colon adenocarcinoma tissues ( = 156), and adjacent normal tissues ( = 124). The expression levels of LGR6 in colon adenoma and adenocarcinoma were significantly higher than those in normal colon epithelial tissues ( <0.001). Low LGR6 expression predicted a short overall survival in patients with colon adenocarcinoma (log-rank test, = 0.016). Univariate and multivariate survival analyses showed that, in addition to N and M classification, LGR6 expression served as an independent prognostic factor. Thus, low expression of LGR6 can be used as an independent prognostic parameter in patients with colon adenocarcinoma.
